چکیده

The average molecular weight of the water soluble hemicelluloses (hemicellulose B) of the Avena coleoptile cell wall was determined by gel permeation chromatography (GPC) and viscometry. Analysis of the neutral sugar composition of henucellulose B eluted from a GPC column (Sepharose 4B) indicated that it consists of /J-glucan with a high molecular weight and arabinoxylan with a low molecular weight. A kinetic study of the effect of auxin on the molecular distribution of henucellulose B demonstrated that auxin decreased the /?-glucan content of the hemicellulose as early as the first hour incubation, but not the arabinoxylan content, when it stimulated the extension of the coleoptile segments. Calculation of the weight-average molecular weight from the chromatograms suggested that auxin decreased the molecular weight of hemicellulose B; this was also confirmed by viscometry. Thus, auxin may cause cell wall loosening, leading to cell extension, through its effect on /?-glucan degradation or through the decrease in the molecular weight of hemicellulose B.

Auxin-induced Changes in Avena Coleoptile Cell Wall Composition.

Sugar and uronic acid residues were derived from wall polysaccharides of oat (Avena sativa, var. Victory) coleoptiles by means of 2 N trifluoroacetic acid, 72% sulfuric acid, or enzymic hydrolysis. The products of hydrolysis were reduced and acetylated to form alditol acetates which were analyzed using gas chromatography. Time-course studies of auxin-promoted changes in various wall fractions i...

Auxin-induced growth of Avena coleoptiles involves two mechanisms with different pH optima.

Although rapid auxin-induced growth of coleoptile sections can persist for at least 18 hours, acid-induced growth lasts for a much shorter period of time. Three theories have been proposed to explain this difference in persistence. To distinguish between these theories, the pH dependence for auxin-induced growth of oat (Avena sativa L.) coleoptiles has been determined early and late in the el...

Inhibition of RNA synthesis and auxin-induced cell wall extensibility and growth by Actinomycin D.

A linear stress strain analyzer was used to determine the effects of inhibitors of RNA and protein synthesis on auxin-induced increases in cell wall extensibility. With etiolated soybean hypocotyl, maize mesocotyl and Avena coleoptile sections and light-grown pea internode sections, inhibition of RNA synthesis resulted in inhibition of auxin-induced extensibility changes and cell expansion. The...

Stoichiometric Correlation of Malate Accumulation with Auxin-dependent K-H Exchange and Growth in Avena Coleoptile Segments.

The action of auxin in the promotion of growth has been suggested in the literature to depend on cell wall acidification. In a former investigation by the present authors the electrochemical balance in auxin-induced proton extrusion was shown to be maintained by potassium net uptake. The present paper reports data demonstrating that the elongation of Avena coleoptile segments is accompanied by ...
